Method Name: Ganciclovir API - Determination of Ganciclovir - Non-Aqueous Titration Method
Scope of application: This method uses titration to determine the content of ganciclovir in ganciclovir raw materials.
This method is applicable to ganciclovir active pharmaceutical ingredient.
Method principle: The test sample is dissolved in glacial acetic acid, then crystal violet indicator solution is added. The solution is titrated with perchloric acid titrant until it turns green. The titration result is corrected with a blank test. Based on the amount of titrant used, the content of ganciclovir is calculated.
Reagent: 1. Glacial acetic acid
2. Perchloric acid titrant (0.1mol/L)
3. Crystal violet indicator solution
4. Potassium hydrogen phthalate as the benchmark
Instruments and equipment:
Sample preparation: 1. Perchloric acid titrant (0.1mol/L)
Preparation: Take 750mL of anhydrous glacial acetic acid (calculated by water content, add 5.22mL of acetic anhydride for every 1g of water), add 8.5mL of perchloric acid (70-72%), shake well, let it cool, add an appropriate amount of anhydrous glacial acetic acid to make up to 1000mL, shake well, and let it stand for 24 hours. If the test sample being tested is prone to acetylation, the moisture content of the solution should be determined by the moisture determination method, and then the moisture content of the solution should be adjusted to 0.01% to 0.2% with water and acetic anhydride.
Calibration: Take approximately 0.16g of the reference potassium hydrogen phthalate that has been dried at 105℃ to a constant weight, accurately weigh it, add 20mL of anhydrous glacial acetic acid to dissolve it, add 1 drop of crystal violet indicator solution, and slowly titrate with this solution until it turns blue. Then, correct the titration result with a blank test. Each 1mL of perchloric acid titrant (0.1mol/L) is equivalent to 20.42mg of potassium hydrogen phthalate. Based on the consumption of this solution and the amount of potassium hydrogen phthalate taken, calculate the concentration of this solution.
2. Crystal violet indicator solution
Take 0.5g of crystal violet and add 100mL of glacial acetic acid to dissolve it.
Operation steps: Accurately weigh approximately 0.15g of the test sample, add 40mL of glacial acetic acid, heat to dissolve, cool, add 1 drop of crystal violet indicator solution, titrate with perchloric acid titrant (0.1mol/L) until the solution turns green, and correct the titration result with a blank test. Each 1mL of perchloric acid titrant (0.1mol/L) is equivalent to 25.52mg of C9H13N5O4.
Note: "Precise weighing" means that the weight taken should be accurate to one-thousandth of the weight being weighed. "Precise measurement" means that the accuracy of measuring the volume should meet the precision requirements for the pipette of that volume as stipulated in the national standards.
